I wonder if there's a limit on file system complexity in iOS. My iPod Touch 64 and iPad 64 seem to be breaking down at the same point, even though my iTunes is the latest and the i-devices have the latest firmware on them. The most common symptom is the video thumbnails all go blank.
The files are identical on each. All manually managed. 2475 MP3's 1999 videos (average about 2 minutes each) 1600 photos 60 apps
About 33 gb used and 25 gb free space. My iPhone 64 has the same files and doesn't have a problem so far.

I changed the hard drive of an iPod 5.5 80 GB to a 240 GB so I could load a lot of Apple Lossless files. The iPod 5.5 with the 240 GB HD now holds over 8,000 (about 204 GB) songs with no problems.

I just completed updating my 64gb iPod, iPad, and iPhone, which were all corrupted with wrong art showing for many songs, most of the video thumbnails missing, various other problems. All were maintained on a very secure company computer so not likely they were corrupted by the PC. Apple's suggestion of deleting various files and reloading didn't work, so there was no point in transferring any settings and risking the restore, so I just restored factory settings. 15 hours work total - 5 hours per device. This has never been necessary for me with a computer, so I suppose I have to consider iOS to be an unreliable O/S.

my iPad2 64gb has over 40gb of music videos on it, and seems fine to me, I also have iPod Classic 80gb and iPod Classic 30gb that have been hammered to death over the last 5 years with no problems, but I don't use iTunes (which I have heard some terrible horror stories with friends getting into all sorts of problems using iTunes over the years)

I can only talk for OSX and iTunes with an iPod Touch 32GB. So far no real problems, but I know that some people had problems.
My iPod Touch 32GB is almost filled up completely, even the Update to iOS 5 was no problem.
I must admit, that I did like iOS 4.3.5 better on the iPod Touch than iOS 5.x.x. The power consumption is greater with iOS 5.x.x and since I don't have a latest generation iPod Touch (I have a 3rd gen), most new features are not really useful for me. Personally, I use the iPod Touch mainly as audio playback device and only a few iApps are important for me (a currency converter for example).
